Nembro lies in the Val Seriana, on the right bank of the Serio. Its name may come from Latin nemus, wood, with an added b, given the forests that covered the area; other guesses derive it from nimbula, cloud, or from Celtic nembren, height. In the early Middle Ages it was, with Clusone, one of the two pieve seats of the valley, and traded with the Val Brembana along the via Mercatorum. The Guelph and Ghibelline quarrels cut it in two: a Ghibelline upper half, fortified on the hill where the church of San Pietro now stands, and a Guelph lower half on the hill of the Zuccarello sanctuary. Decline set in when Venice opened the via Priula, which climbed from Bergamo into the Val Brembana without passing this way.

The town lived on whetstones, quarried and worked in the valley, and later on silk mills built along the Serio beside two old canals, the roggia Serio Grande and the roggia Morlana; a museum in the former casa Bonorandi tells the whetstone story. The Savoldi, brickmakers of Nembro, conceived in the nineteen-twenties the project of the Grande Enciclopedia Italiana, later passed to Treccani. In 2020 Nembro, with Alzano Lombardo, was one of the places where the COVID-19 pandemic began and spread hardest.
